Offer Description
The BOB1-OCT2 transcriptional complex is a lineage-specific master regulator of the germinal center B-cell program, driving both autoimmune pathologies and aggressive B-cell malignancies. Funded by the LABEX IMMUNE from NEXT/Nantes Université, this PhD project aims to synthesize and biologically evaluate small-molecule inhibitors of this protein-protein interaction.
This interdisciplinary consortium involves teams in medicinal chemistry (CEISAM-SYMBIOSE), immunology (CR2TI), and translational onco-hematology (CRCI2NA). The medicinal chemistry component will optimize initial hit molecules by introducing novel azaheterocyclic scaffolds through a scaffold-hopping strategy. Based on robust preliminary results, a synthetic chemistry work package is planned to validate the functionalization of azaheterocyclic structures.
As part of this drug design program, the PhD student will participate in the evaluation of molecules, which will be characterized to determine their efficacy in inhibiting the BOB1/OCT2 interaction and their pharmacological profiles. This series of novel compounds will be evaluated in various in vitro and in vivo models involving B immune cells (autoimmune diseases and cancer).

Interdisciplinary Consortium at Nantes Université with technical supervisions.:
- (CEISAM, Organic & Medicinal Chemistry): Led by Dr Arnaud Tessier providing the medicinal chemistry engine driving rational redesign, scaffold hopping, SAR refinement, and physicochemical optimisation.
- (CR2TI, Team4, Immune regulation): Led by Dr Sophie Brouard, providing deep expertise in B-cell biology and autoimmunity, leading assay development, primary B-cell models, and autoimmune in vivo models.
- (CRCI2NA, Translational Onco-haematology): Led by Dr Catherine Pellat, providing expertise in high-risk B-cell malignancies with advanced functional assays to evaluate oncological mechanisms and biomarker signatures.
